Construction and Application of Recombinant Bacterial Strains Secreting Lactoferrin Peptides

分泌乳铁蛋白肽重组菌的构建及其应用进展

Abstract: Lactoferrin peptides are bioactive small peptides generated through enzymatic hydrolysis of lactoferrin and exhibit multiple biological activities,including antibacterial,antiviral,anti?inflammatory,immunomodulatory, and intestinal barrier protective effects,often showing higher bioactivity than intact lactoferrin. However,their large? scale application is limited by the high cost and low yield of natural extraction. Genetic engineering technology provides an efficient strategy for the scalable production of lactoferrin peptides. This review systematically summarized the construction strategies of recombinant lactoferrin peptide ? producing strains,focusing on different expression systems,including Escherichia coli,lactic acid bacteria,and fungal hosts such as Pichia pastoris,as well as key technical approaches including codon optimization,fusion tags,tandem expression,chromosomal integration,and signal peptide screening to achieve stable and high?level expression. Additionally,the mechanisms of action of these recombinant strains are discussed,where secreted lactoferrin peptides and synergizing with host probiotics,exhibit antibacterial, antiviral, immunomodulatory, and intestinal barrier repair activities. In practical applications, recombinant strains ? secreting lactoferrin peptides have shown beneficial effects on disease resistance,growth performance,and immune enhancement effects in animals such as newborn piglets,weaned piglets and poultry. Furthermore,formulation strategies such as microencapsulation can improve gastric acid tolerance and intestinal colonization capacity,highlighting their potential as promising. With continued advances in genetic engineering and formulation technologies,recombinant lactoferrin peptide?secreting strains are expected to play an important role in animal health and sustainable livestock production.

摘要: 乳铁蛋白肽是乳铁蛋白经酶解产生的小分子活性多肽,具有抗菌、抗病毒、抗炎、免疫调节及肠道屏障保护等多种生物学功能,其活性通常高于完整的乳铁蛋白。然而,提取成本高、产量低的问题限制了其规模化应用,基因工程技术为乳铁蛋白肽的高效生产提供了新途径。该文系统综述了乳铁蛋白肽重组菌的构建策略,包括利用大肠杆菌、乳酸菌及真菌(如毕赤酵母)等多种表达系统,通过密码子优化、融合标签、串联表达、染色体整合及信号肽筛选等技术手段,实现乳铁蛋白肽的高效、稳定表达。同时,阐述了重组菌通过分泌乳铁蛋白肽,协同宿主益生菌作用,发挥抗菌、抗病毒、免疫调节与肠道屏障修复等多重作用机制。在应用方面,分泌乳铁蛋白肽的重组菌在新生仔猪、断奶仔猪和雏鸡等动物生长中表现出良好的抗病、促生长和免疫增强效果,并可通过微胶囊化等制剂技术提升其胃酸耐受性与肠道定植能力,作为饲用抗生素的替代品具有极佳的应用前景。未来,随着基因工程与制剂技术的进一步发展,分泌乳铁蛋白肽的重组菌有望在动物健康养殖中发挥重要作用。
