山羊支原体山羊肺炎亚种和大肠埃希菌混合感染的诊断

Diagnosis of Coinfection of Mycoplasma capricolum subsp. Capripneumoniae and Escherichia coli in goats

摘要: 为确定导致云南省禄丰市某规模化羊场山羊病死亡的致病原,采集病死山羊肺脏组 织及气管分泌物各1份,采用PCR法对主要病原进行鉴定,开展细菌分离、支原体培养、形态学 观察、生化鉴定、药敏试验、测序和遗传进化分析。结果显示,肺脏组织及气管分泌物山羊支原 体山羊肺炎亚种核酸呈阳性;通过肺脏组织成功分离出1株细菌和1株山羊支原体。细菌经生 化鉴定为大肠埃希菌;药敏试验结果显示,该菌对多粘菌素E、卡那霉素、庆大霉素、安普霉素和 呋喃妥因敏感。山羊支原体通过PCR扩增获得670 bp大小的目的基因片段,与NCBI GenBank 上山羊支原体山羊肺炎亚种参考菌株(登录号:CP144022.1)序列同源性达100.00%。结果表 明,引起云南省禄丰市某规模化羊场山羊羊群发病死亡的病因为山羊支原体山羊肺炎亚种与 大肠埃希菌混合感染,该试验结果可为云南本地山羊传染性呼吸道疾病的诊断、治疗和防控提供参考依据。

Abstract: To identify the causative agent responsible for goat deaths on a large?scale farm in Lufeng City,Yunnan Province,lung tissue and tracheal secretion samples were collected from deceased animals. PCR was used for initial pathogen screening, followed by bacterial isolation, mycoplasma culture, morphological observation, biochemical identification,antimicrobial susceptibility testing,sequencing,and phylogenetic analysis. The results showed that samples were positive for Mycoplasma capricolum subsp. Capripneumoniae (Mccp)nucleic acid. One bacterial strain and one Mycoplasma strain were successfully isolated from lung tissue. The bacterial strain was identified biochemically as Escherichia coli,which was sensitive to colistin,kanamycin,gentamicin,apramycin, and nitrofurantoin. The Mycoplasma strain was confirmed by PCR amplification,yielding a a 670 bp target gene fragment with 100.00% sequence identity to the Mccp strain(GenBank accession number:CP144022.1). These findings indicate that the mortality in goats on this farm was caused by mixed infection with Mccp and E. coli. This study provides a reference for the diagnosis,treatment,and control of infectious respiratory diseases in local goats in Yunnan.
