Protective Efficacy of the Chitosan⁃Montmorillonite Intercalation Composite Detoxifier against Deoxynivalenol Exposure in Mice

摘要: 该试验以壳聚糖?蒙脱土插层复合物(Chitosan ? montmorillonite , CS?MMT)为基础, 添加复合益生菌、酶和酵母细胞壁,研发具有吸附和降解呕吐毒素(DON)功能的CS?MMT复合 脱毒剂(CDA)。选择36只5周龄健康雄性C57BL/6J小鼠,随机分为6组(n=6):正常对照组、 DON处理组(2 mg/kg)、CDA单独处理组(100 mg/kg)、CDA低剂量组(50 mg/kg+DON)、CDA中 剂量组(100 mg/kg+DON)以及CDA高剂量组(200 mg/kg+DON)。连续给药21 d并记录体重、 日采食量、每日饮水量以及粪便中DON残留量,试验结束后采集小鼠血清和肠道内容物。结 果发现,CDA处理显著改善了DON暴露小鼠的各项生长性能指标并减少了粪便中DON的残留 量,同时降低了血清中促炎因子的水平。此外,CDA通过多效方式增加肠道菌群的丰富度,促 进有益菌的生长并抑制有害菌的繁殖。CDA能够改善DON暴露小鼠的肠道健康,为缓解DON 污染对动物健康的危害提供了新的思路。

Abstract: This experiment was based on the chitosan ? montmorillonite intercalation composite (Chitosan ? montmorillonite,CS ? MMT),and incorporated probiotics,enzymes and yeast cell walls to develop a CS ? MMT composite detoxification agent(CDA)with the functions of adsorption and degradation of deoxynivalenole. Thirty?six healthy male C57BL/6J mice at 5 weeks of age were selected and randomly divided into 6 groups(n=6):normal control group,DON treatment group(2 mg/kg),CDA alone treatment group(100 mg/kg),CDA low?dose group (50 mg/kg + DON),CDA medium?dose group(100 mg/kg + DON),and CDA high?dose group(200 mg/kg + DON). The mice were given continuous treatment for 21 days and their body weight,daily feed intake,daily water consumption,and DON residue in feces were recorded. After the experiment,blood serum and intestinal contents of the mice were collected. It was found that CDA treatment significantly improved various growth performance indicators of DON?exposed mice and the residual amount of DON in their feces,and reduced the levels of pro?inflammatory factors in their serum. Moreover, CDA increased the richness of the intestinal microbiota through multiple mechanisms,promoted the growth of beneficial bacteria and inhibited the reproduction of harmful bacteria. CDA can improve the intestinal health of DON?exposed animals,providing a new approach to alleviating the harm caused by DON contamination to animal health.
